Forces and stresses in epithelial monolayers

The mechanical behavior of living tissues is deeply connected with many important biophysical questions, yet little is known about internal tissue mechanics. Since the traction forces exerted by cells on a planar, deformable substrate can be measured, we combine traction force data with Bayesian inversion to estimate the internal stress field of a cell epithelial monolayer. The method is validated using numerical simulations as well as experimental data. Importantly, its accuracy does not depend on the rheology of the tissue. Given traction force timelapse movies, we show that the tissue stress field can also be estimated by Kalman filtering. Inferred stresses allow to quantify the rheology of epithelial cell sheets in physiological conditions. In a tissue where cell rearrangements are rare, we find that cell shape anisotropy is a good proxy for deviatoric stress.
