Mechanics and polarity in B cells

Spatio temporal dynamics of immune cell, Inserm U932, Institut Curie

B cells are the antibody producing cells of immune system. To mount an efficient immune response, B cells recognize and internalize the antigen through a specific signaling platform called immune synapse. The formation of such a structure entails force generation and polarization of cellular structures that we have studied in details in the past years. Recently, we used functionalized microdroplets as presenting-cell models and a microfluidic pairing device to observe multiple synchronized synapses. This system allowed us to understand the role of the different physico-chemical properties of the presenting surface in polarity. We also identified an early and a late phases in polarity dominated, respectively, by actin and microtubules dynamics and further highlight an unexpected role for microtubules in restricting F-actin polymerization to allow the formation and maintenance of a unique immune synapse.
