Mitochondrial dynamics as a new mechanics-to-biology transduction module

Abstract : Cells have the remarkable ability to measure the spatial and mechanical constraints imposed by their surroundings. Sensing of ECM elasticity depends on a reciprocal feedback between resisting forces exerted by the ECM, transmission of forces across cell-ECM adhesions, and cell-based tension of the actomyosin cytoskeleton attached to adhesions. In response to such constraints, cells regulates complex behaviors including the switch from proliferation to quiescence, cell fate differentiation or death, and the acquisition of distinct metabolic profiles, which bears relevance in the context of tissue homeostasis and pathology. Our recent work indicates that extracellular physical constraints reverberate onto intracellular organelle dynamics to regulate these phenotypes, and especially to coordinate key metabolic traits that empower cell fitness and resistance to oxidative stress.
