Mechanics of membrane nanotubes and micropatterned cancer cells probed by AFM

Abstract : Our lab studies the relationship between biological functions and the mechanics of living objects. I will show results from two ongoing projects, one on living cells and one on reconstituted biomimetic systems, both using AFM nanomechanical mapping. First, I will show how AFM allows assessing the morphology and mechanics of cancer cells cultured on adhesive micropatterns in relation to their metastatic potential, as an attempt to standardize AFM mechanical phenotyping. Then, I will present results on the mechanics of biomimetic membrane nanotubes, structures commonly involved in intracellular trafficking. A theoretical description of the AFM tip-membrane interaction allows us to relate AFM measurements of the nanotubes’ morphology to the membrane mechanical parameters. In addition, we use AFM to assess the induced changes in nanotube physical properties when actin polymerizes at their surface. We show that this approach can be extended to phase-separated membrane nanotubes.
