Probing Red Blood Cell stiffness with fluorescent probes : towards new markers of pathology

Abstract : We propose a new method for intracellular rheology of red blood cells (RBCs) based on molecular rotors, fluorescent viscosity probes. The relevance of the technique is established on healthy RBCs whose stiffness varies with temperature or between donors. We compare the sensitivity of this new method with the reference technique for deformability measurements in erythrocyte pathologies, ektacytometry. In the case of sickle cell disease, we propose new indicators that could complement the clinical reference methods.
