Biofilament Confotronics : Hidden Internal Dynamics of Actin Filaments and Microtubules

Microtubules (MTs) and actin filaments (AFs) are dominant structural components of the eukaryotic cytoskeleton. Their interesting polymerization behavior and interaction with motors have been central themes of biophysical investigations in the past. However it is far less known that even in their stabilized form (and in absence of motors) MTs and AFs exhibit very interesting internal lattice dynamics. This internal dynamics seems related to yet unidentified polymorphic lattice switching events. We will present fresh experimental evidence for torsional AF switching and long range cooperative polymorphic transitions of stabilized MTs and introduce theoretical tools for their understanding. The hidden polymorphism can dramatically affect the filament’s elastic behavior as most strikingly illustrated by supercoiling of AF rings and dramatic straight to curved transitions of single MTs. We end with a thought provoking conclusion : Microtubules could act as "conformational wires" and rapidly transmit structural information on sub-second time scales along their full contour on typical cellular length scales !
