Studying cytoskeleton symmetry breakings and signaling concentration gradients using magnetic nanoparticles

We will discuss two aspects :

(1) The role of a geometrical confinement on the self-organization of microtubules and molecular motors : We have developed an in vitro system, based on the cellular reconstitution by confinement of Xenopus egg extracts, to examine simple aspects of symmetry breakings of the cytoskeleton.

(2) The role of a spatial asymmetry in biochemical signals in microtubule organization : Intracellular biochemical reactions are often localized in space and time, inducing gradients of enzymatic activity that may play decisive roles in determining cell’s fate and functions. However, the techniques available to examine such enzymatic gradients of activity remain limited. Here, we will discuss a method to artificially control in space and time such signaling pathways using magnetic nanoparticles. For instance, this approach allows to examine the role of reaction-diffusion processes and concentration gradients of GTPase signaling proteins on microtubule morphogenetic properties.
