Forces, pressure and spontaneous curvature in Yeast endocytosis
Endocytosis is a process by which external molecules are internalized into vesicles by the invagination and severing of the plasma membrane. Several interconnected biochemical pathways have been identified the last decades, however the physical mechanisms controlling the deformation and cutting of the membrane remain elusive. For instance, the exact role of coat proteins (such as Dynamin and Rvs167) is heavily discussed.
Combining theoretical modeling of the membrane mechanics and recent high resolution experimental data, we show the fundamental role of the turgor pressure in opposing membrane deformation in Yeast cells. We show that actin organizes in a rigorous fashion prior to membrane deformation to exert large pulling forces generated by polymerisation, and that the membrane rigidity is much larger than expected. Additionally, we show that the aggregation of coat proteins along the membrane surface yields a shape instability able to sever the membrane invagination.
