Spatial control of LBL for musculoskeletal tissue engineering

In vivo, cells are embedded in a microenvironment that contains several extracellular matrix proteins, growth factors and also mechanical signals from the tissue and from neighboring cells . All these biochemical, mechanical and topographical signals are important to orient cell fate. In this presentation, I will present our recent studies on layer-by-layer films made of polysaccharides and polypeptides applied to musculo-skeletal tissue engineering. We aim to mimic some aspects of the cell microenvironment, especially controlled bioactivity with presentation of bioactive peptides and growth factors, mechanical properties, and topographical properties to guide muscle cell spatial organization and fate. Skeletal precursor can differentiate either into mature myotubes or into bone cells in the presence of osteo-inductive proteins, such as BMP (bone morphogenetic proteins). Combination of microstructured surfaces, porous scaffolds, LbL films and microtechniques enable to study tissue formation in well defined microenvironments, both in vitro to answer fundamental biological questions and in vivo to improve osteointegration of orthopaedic implants.
