The morphology of mature biofilms have been reported to adapt to environmental challenges. Yet, if some proteins influencing the shape have been identified, mechanisms by which those shapes are modulated are barely known. In particular, early steps of biofilm formation are critical as bacteria have to proliferate while maintaining contact on the surface they colonize. Using timelapse force microscopy of E. coli and P. aeruginosa microcolonies, we show that adhesions are heterogeneous and dynamical. Moreover, at the single cell level, adhesions tend to localize at the old pole of cells. Experiments using adhesion mutant strains as well as simulations show that the cellular localization of adhesions as well as their strength determine the shape of two-dimensional microcolonies and trigger their transition to a three-dimensional growth regime. This study propose the interaction with the surface as a major levier by which rod-shape bacteria module the shape of their group.
