Cells must control their volumes to maintain high levels of macromolecular crowding. In bacteria and other microorganisms, cell volume is set by the peptidoglycan (PG) cell wall. Using Escherichia coli as a model organism we monitored cellular dry mass and cell dimensions during growth at the single-cell level using quantitative phase microscopy. We found that dry-mass density is well-conserved during steady-state growth and during shifts of growth conditions. We found strong evidence that cells robustly control cell volume by coupling the rate of surface-area increase linearly to mass growth and by fine-tuning cell diameter. Cell length, on the contrary, is a simple consequence of the control of surface area and diameter. Interestingly, the rate of cell-surface expansion is independent of cell-wall synthesis, contrary to the current paradigm. Proper cell diameter, on the contrary, strongly depends on cell-wall synthesis. If time permits, I will present our work on the molecular and physical determinants of cell diameter and straight cell morphology through cell-wall synthesis.
